I'm the Juggernaut!
In the second volume of Pokespecial, Red discovers the unfinished project of Mewtwo, then when he talks to Blue, she asks Red if he knows how many pokemon there are. He responds 150, when Blue frantically exclaims 151.

Now my question is, if Mewtwo was the 150th Pokemon, and at that time it wasn't completed, why did Red say 150? Was it just a careless mistake?

Well, statements regarding the actual number of pokemon species is always tricky in any medium. I suppose you can say it's a mistake, and the writers were probably too intent in mimicking the game facts that only 150 pokemon species were confirmed at that time, and Mew was supposed to be the additional one that no one has heard of.

But then again, in terms of plothole, Blue would certainly have seen more than 150 herself since she grew up with the Mask of Ice, and the 'Escape!' chapter already showed that she has at least seen Sneasel, Natu, Hounder, and of course her own Snubbull which she traded to Silver.

In conclusion, just regarding the RGB series alone, yes, that accounts for a mistake I guess, since the mentioned 150 was clearly intended to refer to the Kanto 150, which includes Mewtwo.
